Note that your active Gcode window shows G3. So you are in ARC mode and by starting at line 40010 you are telling it to move to those coordinates in an arc motion which does not work. Line 40010 is supposed to be a G0 rapid move.
This is the problem with "start from line". You need to make sure all the modal G codes are set correctly for the move.
